A new study on the use of sildenafil, marketed as Viagra, has found the medication could cause a small, potentially permanent (up to 1/10,000,000) case of Alzheimer's disease.
A small number of patients have suffered from an Alzheimer's disease-causing condition, but researchers have found that the drug may be effective in reducing the risk of dementia.
Researchers examined records of more than 300,000 patients who had taken sildenafil for at least four years. The drug was taken in pill form by mouth.
Sildenafil was taken at a dose of 50 milligrams (mg) twice daily. The researchers found that patients with an average age of 65 years had a greater risk of developing dementia than patients who were younger, with a risk of 16 per cent versus only 1 per cent in the older group.
About 2.3 million people in the United States took the drug in 1998.
The study is the first to quantify the risk of dementia, or Alzheimer's, in patients taking sildenafil for a year.
The study showed that the drug was not associated with a significant increase in the risk of dementia, compared with those taking a placebo. The risk of developing dementia was not increased by the dose of sildenafil taken.
Dr. John Smith, director of the Alzheimer's Disease Research Centre, said: "These results are encouraging and will likely be of benefit to those already receiving this medication, particularly those with other risk factors for dementia, and who may be at increased risk for developing dementia."
"We are encouraged by the findings, and we are looking for more information on this potential cause of dementia and the potential benefits of sildenafil for many."
Sildenafil is marketed under the brand name Viagra. It is sold under the brand name Revatio.
The drug was taken by mouth, about two hours before the patients were expected to take the medication. Doctors have only occasionally found that the medication could increase the risk of dementia in patients taking sildenafil.
The researchers found that the drug was not associated with a significant increase in the risk of dementia.
Patients who took sildenafil daily had the greatest risk of developing dementia compared to patients who did not take the medication.
However, the researchers say the findings suggest that sildenafil may have a beneficial effect in preventing dementia in some people.
"It is important to note that the results of the study do not mean that all patients will have the benefit of sildenafil," Smith said.
The study was funded by Pfizer Inc. and was published in the journal Alzheimer's Disease. The journal, which is not part of the research, was not involved in the study.
